Nupr1/Chop Signal Axis Is Involved in Mitochondrion-Related Endothelial Cell Apoptosis Induced by Methamphetamine
Researchers identified a novel pathway involved in endothelial cell apoptosis induced by methamphetamine (METH). They demonstrated that exposure to METH caused mitochondrial apoptosis in human umbilical vein endothelial cells and rat cardiac microvascular endothelial cells in vitro as well as in rat cardiac endothelial cells in vivo. VEGF Receptor-2-Specific Signaling Mediated by VEGF-E Induces Hemangioma-Like Lesions in Normal and in Malignant Tissue
Researchers generated transgenic mice specifically expressing ovVEGF-E in β-cells of the endocrine pancreas (Rip1VEGF-E; RVE). RVE mice showed an increase in number and size of the islets of Langerhans and a distorted organization of insulin and glucagon-expressing cells. Islet endothelial cells of RVE mice hyper-proliferate and form increased numbers of functional blood vessels. [Angiogenesis] Abstract

A Mutation in SLC22A4 Encoding an Organic Cation Transporter Expressed in the Cochlea Strial Endothelium Causes Human Recessive Non-Syndromic Hearing Loss DFNB60
Using immunofluorescence, scientists found that Slc22a4 is expressed in stria vascularis (SV) endothelial cells of rodent cochlea and targets their apical plasma membrane. They also found Slc22a4 transcripts in our RNA-seq library from purified primary culture of mouse SV endothelial cells. [Hum Genet] Abstract

Adipose-Derived Stem Cells Support Lymphangiogenic Parameters In Vitro
Scientists have conducted co-culture experiments to investigate the effects of human adipose-derived stem cells on human lymphatic endothelial cells in terms of gene expression profile, proliferation, migration, and tube formation in vitro. [J Cell Biochem] Abstract

Critical Endothelial Regulation by LRP5 during Retinal Vascular Development
Researchers showed that loss of LRP5 function in mice causes retinal hypovascularization during development as well as retinal neovascularization in adulthood with disorganized and leaky vessels. Using a highly specific Flk1-CreBreier line for vascular endothelial cells, together with several genetic models, they demonstrated that loss of endothelium-derived LRP5 recapitulates the retinal vascular defects in Lrp5-/- mice. ER Alpha Rapid Signaling Is Required for Estrogen Induced Proliferation and Migration of Vascular Endothelial Cells
Researchers found that in endothelial cells (ECs) expressing wild type estrogen receptor (ER) alpha, the active endogenous form of estrogen regulates many genes involved in cell migration and proliferation, promotes EC migration and proliferation, and also blocks the adhesion of monocytes to ECs. Inhibition of p38 Mitogen-Activated Protein Kinase Enhances the Apoptosis Induced by Oxidized Low-Density Lipoprotein in Endothelial Progenitor Cells
Investigators showed that oxidized low-density lipoprotein (oxLDL)-induced p38 phosphorylation in endothelial progenitor cells is time and dose dependent. Treatment with antioxidant N-acetyl cysteine restored oxLDL-induced p38 phosphorylation to basal levels. [J Vasc Res] Abstract

Syndecan-4 Shedding Impairs Macrovascular Angiogenesis in Diabetes Mellitus
Researchers investigated the alterations of syndecan-4 (synd4) endothelial expression in diabetes mellitus and the influence of impaired synd4 signaling on angiogenesis in human umbilical vein endothelial cells, diabetic rats, synd4 null mice, and db/db mice. [Biochem Biophys Res Commun] Abstract

Tirofiban Induces Vasorelaxation of the Coronary Artery via an Endothelium-Dependent NO-cGMP Signaling by Activating the PI3K/Akt/eNOS Pathway
Investigators found that tirofiban induces endothelium-dependent vasodilation in a concentration- and time- dependent manner in the isolated rat coronary artery pre-constricted by 5-hydroxytryptamine. They also showed that incubation of human umbilical venous endothelial cells with tirofiban increased NO production, which is ascribed to the increased eNOS phosphorylation.
